چکیده
Experimental allergic encephalomyelitis is produced by injection of a basic protein from centralnervous tissue mixed with Freund's complete adjuvant into guinea pigs (Kies, 1965; Carnegie, Bencina & Lamoureux, 1967). The encephalitogenic basic protein is a component of myelin (Laatsch, Kies, Gordon & Alvord, 1962; Roboz-Einstein, Csejtey & Davis, 1965; Eng, Chao, Gerstl, Pratt & Tavaststjerna, 1968). Huuman brain was defatted with chloroformmethanol (2:1, v/v) and the encephalitogenic protein extracted at pH2-5-3-0. The basic proteins were collected on CM-Sephadex C-25 and eluted with 081N-HCI. Gel filtration on Sephadex G-100, G-75 and G-50 was used to purify the encephalitogenic protein. In calculating the amino acid composition shown in Table 1 the protein was assumed to contain six histidine residues, since six histidyl peptides were observed in a peptide 'map' of a tryptic digest. The values for serine, threonine and tyrosine are extrapolated to zero time of hydrolysis with constant-boiling HCI. Tryptophan

Designing, Optimization and Construction of Myelin Basic Protein Coding Sequence Binding to the Immunogenic Subunit of Cholera Toxin
Abstract Background and Objectives: Multiple sclerosis (MS) is a chronic inflammatory autoimmune disease. Mucosal feeding of myelin basic protein binding to the cholera toxin B subunit can reduce the intensity of the immune response in MS patients. Expression system, the domain composition of the fusion protein, accessibility of two domains, codon adaptation index (CAI) and GC contents are v...
متن کاملAllergic encephalomyelitis. Isolation of an encephalitogenic peptide active in the monkey.
A 17-residue peptide (Peptide Y) was isolated from the COOH-terminal end of the basic protein of bovine myelin by peptic digestion. This peptide induced experimental allergic encephalomyelitis in the rhesus monkey. Treatment of Peptide Y with cyanogen bromide released three amino acids from the COOH-terminal end and resulted in a tetradecapeptide (Peptide M) which was also encephalitogenic in t...
متن کاملFusion and sequence analysis of the influenza A (H9N2) virus M2e and C-terminal fragment of Mycobacterium tuberculosis HSP70 (H37Rv)
The present study was aimed to construct a fusion plasmid harboring the extracellular domain of the influenza A M2-protein (M2e), which was fused to the N-terminus of the truncated HSP70 (HSP70359–610) molecule as a new approach for future vaccine research against influenza A. The amplified fragments, M2e and HSP70359-610 genes, were gel-purified. The products were then single digested with Bam...
متن کاملAmino acid sequence of the basic protein of the myelin membrane.
The amino acid sequences of the encephalitogenic basic protein, A1, from bovine and human myelin are similar, differing by only 11 residues. The sequence reveals that while basic residues are spread randomly over most of the polypeptide chain, several regions (8-10 residues) exist that are nonpolar in character. The bovine protein has 170 residues with molecular weight 18,400. The human protein...
متن کاملAntifungal Activity of Heterologous Expressed Chitinase 42 (Chit42) from Trichoderma atroviride PTCC5220
The cDNA from the mycoparasitic fungus Trichoderma atroviride PTCC5220 encoding a 42 kDa chitinase (Chit42) was isolated. The nucleotide sequence of the cDNA fragment as having a 1263 bp open reading frame that encodes a 421 amino acid polypeptide, and a high homology was found withother reported Chit42 belonging to the Trichoderma sp. The 22 amino acid N-terminal sequence is a putative s...
